Objective:To study the effect of Butylphthalide on Intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) in rat with middle cerebral artery ischemia-reperfusion. Explore the Neuroprotective effects of butylphthalide.Methods:Transient middle cerebral artery occlusion(tMCAO)rat models were established by using Longa’S suture insertion.40 health male SD rats were selected and randomly assigned into 4 Groups:control group(N), sham-operated grop(S),butylphthalide pretreatment group(I/R) and Ischemia-reperfusion groups(D I/R),each group had 10 rats. The rats of butylphthalide pretreatment group received intraperitoneal injection of 3ml edaravone evey day before the molding, and the others rats received intraperitoneal injection of 3ml physiological salin evey day before the molding.24 hours later, the brain and serum were collected and their expression of VCAM-1 and ICAM-1 was detected using ELISA and immunohistochemistry. All datas in the groups were analyzed with t test.Results:1.No neurologic impairment was observed in the rats of control group and sham-operated grop, while NDS of butylphthalide pretreatment group and Ischemia-reperfusion groups were higher than control group and sham-operated grop; Compared to schemia-reperfusion group, NDS was significantly lower in butylphthalide pretreatment group (P<0.05); 2.The expression of ICAM-1 and VCAM-1 in the butylphthalide pretreatment group and Ischemia-reperfusion groups were higher than control group and sham-operated grop at reperfusion 24 hours (P<0.05). while VCAM-1 expression was higher in Butylphthalide pretreatment group compared to the Ischemia-reperfusion group (P<0.05)。Conclusion:1. High expressions of ICAM-1 and VCAM-1 may play a key role in the Cereral isehemia-reperfusion iniury.2. Neuroprotectionve may reduce the brain damage after cerebral isehemia and has neuroprotective effects on brain.
