Objective:By making type 2 diabetic rat models to observe changes of pancreatic blood flow and to study the affection of high blood glucose to pancreatic microcirculation and explore the pathophysiological mechanism of pancreas with increasing blood glucose, in order to make reference for clinical prevent and treatment of microcirculation disorder in Type 2 Diabetic duration.Methods: Randomly divided 50 Wistar rats of 7-week old weighting 160~180g into two groups, high-fat fed streptozotocin-diabetic group(experiment group) and normal normally fed group(control group),the former group contains 45 Wistar rats, the latter contains 10 rats. Experiment group experiment group were fed with high-fat foods for 4 weeks. The rats were fasted 8~14 hours, then the blood glucose , insulin and islet blood flow were determined. The high-fat fed streptozotocin-diabetic rats were given STZ 30mg/Kg i.p. once a week, OGTT was made after 2 weeks. Those rats with FPG≥7.1mmol/L or 2HPG≥11.1mmol/L were regarded as successful models.
