[Objective]To research the survival, migration and differentiation of neural stem cells (NSCs) in the host animals following transplanted into the adult rats striatum, providing evidence for NSCs transplantation; research the effect of NSCs transplantation on symptoms of Parkinson's disease animals.[Methods]The research work starts from dissociation and culture of E14 rat subventricular zone (SVZ) neural stem cells, according to the methods of neurophere culture, a great deal of neurospheres were obtained. To determine the phenotype of cultured cells, the cells were separately immuno-stained with anti-Nestin antibody, anti-NF antibody, anti-GFAP antibody, or anti-CNP antibody.After 3 passages, some NSCs were infected by adeno-associated virus vectors containing the marker gene-green fluorescent protein (AAV2-GFP), GFP labeled NSCs were transplanted into adult Sprague-Dawley rat striatum. The animals were allowed to survive for 45days, 90days or 120days before they were purfused. All the fixed brains were sectioned in the saggital plane, immunohistochemical protocols and fluorescent microscopes were both used to assess the survival, migration and differentiation of transplanted NSCs.6-hydroxy dopamine (6-OHDA) was injected into medial forebrain bundle (MFB) of adult SD rats to make parkinson's disease (PD) animal models. One month later, apomophine was injected into the animals abdominal cavity to induce the rats' rotation towards one side. The rats which met behavioral requirements were considered as successful PD animal models. A number of neural stem cells were transplanted into the successful PD animal models' striatum, then experiments were carried out to assess effect of NSCs transplantation on symptoms of Parkinson's disease animals.[Results](1) The neural cells dissociated from E14 rat subventricular zone had the capacity to...
