In the long period, people always think that the regeneration ofneurons of the mammal central nervous system (CNS) is constricted inthe time of embryonic and early postnatal period. That is to say, theneurons located in the brains and spinal cords of adult mammals canonly reduce gradually but not to be refreshed and substituted. However,the regeneration phenomenon of the CNS and the finding of neural stemcells bearing the ability of reproduction challenge the theoryaforementioned. Anterior subventricular zone (SVZa) is a major area,which can produce neural stem cells (NSCs). Studies have shown that,after the NSCs produced in the SVZa, they can migrate to the olfactorybulb (OB) in a highly circumscribed tunnel, which we called rostralmigratory stream (RMS). And when these NSCs arrived at the OB, theykeep moving to the adjacent granular cells and pericytes, theirdestination respectively, to differentiate to granular cells andpericytes. In this study, we adopted immunohistochemistry the learnthe mechanism of how the trauma of decortex vesseles prompt the SVZneural progenitor cells and glial progenitor cells to proliferate anddifferentiate into new neural cells, which formed some migrationpathway to the pars affecta to participate the repairation.1 Effects of decortex vessels on the NSCs located on the adult rat SVZand the role of Tongluojiunao Injection In this study, we use adult male Wistar rats, remove the left cortexvessels and adopt the immunohistochemistry (IHC) methods to explorethe effects of decortex vessels on the proliferation, differentiationand migration of the SVZ NSCs, and in the meantime, the role of theTongluojiunao Injection. Through the IHC results of serial sections,we see that after the decortex operation, the numbers of PCNA, Vimentinand GFAP positive cells increased greatly. And their positive responsesand morphologic changes are striking. Such trauma stimulates therelative static SVZ NSCs to proliferate and differentiate. After theadministration of Tongluojiunao Injection, we can see the increasingand strengthen of the expression of each positive cell. From this wecan make a speculation that Tongluojiunao Injection can prompt the SVZNSCs of decortex rats to proliferate and differentiate throughup-regulate the endogenous nerve growth factors.2 Decortex vessel in adult rats induce the subventricular zone neuronsto proliferate and migrate to the pars affecta In our study we find out that: decortex vessels in adult rats willnot only promote the NSCs in the anterior horn of SVZ and RMS and alsothe inferior horn to proliferate and form a migration stream to thelesion part through corpus callosum. H chain is start from the up wallof inferior horn; L chain from the lateral wall of inferior horn;in the same time, A chain from medial wall of lateral ventricle. Theingredients of those chains include GFAP positive mature astrocytesand Vimentin positive immature astrocytes; Both of them migrate tothe lesion site to attend the regeneration and repairing work.3 Effects of decortex vessels on the NSCs located on the injured siteof adult rat and the role of Tongluojiunao Injection The result of our study has shown us that after the decortex vesselsoperation, the PCNA, GFAP and Vimentin positive cells located at SVZ,through the RMS pathway or enter the corpus callosum (CC), form aapparent migration stream to the injured site. And also, the numberof positive cells is increased. This indicates that it has thepossibility that those NSCs could substitute the damaged neurons andreform the synapse connection to recover the normal function. After the administration of Tongluojiunao Injectia, we can see thenumber of each positive cell is increasing. And the morphologic changesare even more substantial. This result tell us that TongluojiunaoInjectia can prompt the SVZ NSCs, through up-regulate the endogenousnerve growth factors, can proliferate, differentiate and migrate tothe lesion site to take the place of those neurons which are in theprocess of necrosis and apoptosis.
