Objective:The research on the therapy of CNSP and rehabilitation affects proliferation, migration and differentiation of endogenous neural stem cells of rats after cerebral infarction.Methods:Injection Dil fluorescent dyes to the male Wistar rats intracerebroventricular,the aims pre-tag subependymal cell.After the injection,we adopt stochastic method to group the rats.After injection 48h,the animal model was made by blocking middle cerebral artery; we inject BrdU to the postoperative rats in different points,then we use rehabilitation and CNSP with rehabilitation to treat the different groups.All mice were executed after 21d, and the research samples were got at the same time.We selected neuronal function evaluation,halter test,rotation test,retieular screen test,insitu hybridization andimmunofluoresence staining as the indexes.After the wounds,comparative observation of the result between the groups shows after 24h,7d,14d,21d.We were trying to show the effection on the therapy of CNSP and rehabilitation regulated proliferation,migration and differentiation of endogenous neural stem cells of rats after cerebral infarction from the neurological scores,behavior medieine scopes,insitu hybridization result and immunofluoresence staining resultResult:1.Neuronal function evaluation of rats in every group had no obvious difference before therapy.The improvement of the CNSP and rehabilitation point group was significant(P<0.01),the difference in comparison with the rehabilitation group were remarkable:(P<0.05)2.Behavior medieine scopes of rats in every group had no obvious difference before therapy and 24h.After 7d,14d,21d,the improvement of CNSP and rehabilitation group was significant(P<0.01),the difference in comparison with the rehabilitation were remarkable: (P<0.05)3.The therapy of CNSP and rehabilitation promoted the expression of FGF-2mRNA and NCAM in neuron cytoplasm,glial cell cytoplasm,endothelium,positive cell counting has a significant difference compared with the two others group(P<0.01).4.The therapy of CNSP and rehabilitation promoted subependymal zone cell marked by Dil migration,differentiation.We observed Dil/BrdU/NeuN or Dil/BrdU/ GFAP—positive cells in the CNSP and rehabilitation group increasing significantly,compared with two others group(P<0.01).5.Expression of BrdU/NeuN and BrdU/GFAP-positive cells in the CNSP and rehabilitation group more than two other groups,there are a significant increase compared with two others group(P<0.01).Conclusion:1.The therapy of CNSP and rehabilitation promoted rehabilitation of neuronal function after focal ischemia,and it was better the therapy of simple rehabilitation.2.The therapy of CNSP and rehabilitation improved balance,walking and grasping ability of cerebral infarction in rats,increased thigh muscle strength,and it was better the therapy of simple rehabilitation.3.The therapy of CNSP and rehabilitation promoted NSC in neuroproliferation center-SVZ proliferation after focal ischemia,to reduce the diminishment of proliferation with the time passed.4.The therapy of CNSP and rehabilitation promoted that Dil-label cells in SVZ migrate to striatum and cortex of peri-infarct region and differentiate into neurons and astrocytes.5.The therapy of CNSP and rehabilitation increased the expression of FGF-2 and NCAM in focal ischemia brain tissue,and extend expressive time,and it was better the therapy of simple rehabilitation.6.The cluster needling of scalp point therapy can promote the migration of SVZ NSC after focal ischemia.BrdU/NeuN and BrdU/GFAP positive cells expression in the cluster needling of scalp point group are more than two other groups,this therapy can promote the differentiation of endogenous neural stem cell,and it was better the therapy of simple rehabilitation.
