Burn patients often encounter many problems which blocking the management. But the key problem is burn wound healing. During the wound healing, wound dressing change was inevitably. Theoretically, external application of some agents on the burn wound would be benefit for better and quicker wound healing with different acting mechanism. Among the complicated process of burn wound healing, the proliferation and migration of the fibroblasts and epithelia are two important aspects. Fibroblasts are main part of granulation tissue. Which could synthesize and secrete some extracellular matrix such as collagen, fibrin and hyaluronic acid as well as many cytokines which would participate wound healing. The immigration, proliferation and differentiation of the epithelia would determine the wound re-epithelization. For the present, the category and the dosage form of some clinical external application reagents covered wide range. And the selection and application of external agents exhibited extremely irregular and ad libitum. This in big degree interfered with the clinical results of treatment. In order to further explore the influence of some external application aregents and rhGH on the proliferation of fibroblasts and epithelia and to investigate the dose-effect relationship between the external application reagents and rhGH and fibroblasts and epithelia, the fibroblasts and Hacat were cultured in vitro and were acted by some different kinds external application reagents in different concentrations. The changes of the biological characteristics of these cells were observed, so as for us to provide theoretical reference for the logical selection of external application reagents for burn wound.Objective:1. To establish the primary fibroblast culture and to accomplish the passage culture of fibroblasts and Hacat. Some external application reagents and rhGH were employed to stimulate the above cells.
