Objective:The clinical requirement for platelets is increasing rapidly because platelet transfusion has been an important measure for hemorrhage with thrombocytopenia, but the clinical application of fresh platelet concentrate is limited due to insufficient resources, short shelf life at room temperature and a contaminated tendency. Cryopreservation for platelets makes it possible to reserve adequate and longer shelf life platelets according to the clinic requirement. Therefore, the home and overseas scholars pay an attention to the storage lesion of the cryopreserved platelet extensively. Because the mechanism of the storage lesion is concerned with the activation and the metabolic changes of platelet,many scholars have been carried out the studies on the storage lesion of the cryopreserved platelet and the activation. P-selectin (CD62p), which is an actived marker to express on the surface of platelet membrane, is known as an index of observation. The studies on the storage lesion of the cryopreserved platelet and the metabolic changes also have been carried out. Phosphatide to express on the surface of platelets is considered as an index of observation. Whereas,there is shortage of the study on dynamic changes of the storage lesion of the cryopreserved platelet during the whole shelf life. For this reason, the cryopreserved platelets from single-donor during the shelf life within four weeks were the object of the study, CD62p and phosphatidalserine (PS) were the index of the storage lesion of the cryopreserved platelet, 0h, 24h, 72h, 1w, 2w, 3w and 4w after the cryopreservation were the time point of the observation respectively, we used flow cytometry to detect the expression of CD62p and Annexin V on the surface of platelet membrane, combining with the parameters such as...
