1 Background and AimTransforming growth factor β1 ( TGF-β1) is a potent immunosuppressivecytokine that has a pivotal role in tumorigenesis ,development and metastasis. TGF-β1 expressed in almost all solid tumors of meso-,endo- and ectoblastic origin and in tumor cell lines of hematopoietic origin.Active TGF-pi is disulfide-linked homodimmers of two 112-amino acid and 12.5kDa-molecular weight chains.TGF-β1 signaling mediated by specific receptors on cell membrane.Investigation results demonstrated that tumor cells of overexpressing TGF-β1 become refractory to TGF-β1-mediated growth arrest in the late stage of tumor, either due to loss of TGF-βs receptors or due to dysregulation in TGF-βs signaling pathways.Overexpressed TGF-β1 of tumor cells mainly act on immunocytes and stroma cells, that play an important role to promote tumor growth and metastasis. Neutralization of TGF-β1 or inhibition of its production is an effective method of cancer treatment in variety of animal models.Nowadays,TGF-β1 inhibitors are in research, such as antisensenucleic acids inhibiting the synthesis of TGF-β1, small molecule inhibitors (Tranilast) blocking the secretion of TGF-β1,...
