Objective: Edaravone is the first free radical scavenger developed by Mitsubishi company in Japan for cerebral infarction. It is an active anti-oxidizer to capture hydroxy-free radicle. As an new drug for acute cerebrovascular disease, Edaravone is evaluated its protective effect through observing change of the cerebral infarction area in rats cerebral ischemia-reperfusion and expression of NF- κ B, Caspase-3 in this experiment. Nuclear factor - κ B is an important transcription factor and the key step of inflammatory medium. Caspase-3 is a performing protein in apoptosis and the center in apoptosis regulatory program. The activation of Caspase-3 means the death of cell, so it is called apoptosis protein. By observing the change of NF- κ B and Caspase-3 after using of Edaravone for cerebral ischemia-reperfusion in rats, free radical scavenger is further learned the effect on Caspase-3 and NF- κ BMethods: 66 wistor rats were randomly divided into 3 groups, sham operation group, saline control group and Edaravone treatment group. Saline control group and Edaravone treatment group were divided into 6h, 24h, 48h. The left middle cerebral artery ischemia-reperfusion injury model (MCAO) was made by Koizumi suture-occluded method. Neurological function grade was used to examine the behavioral recovery on the same rats. TTC staining was used to calculate the cerebral infarction area. The concentration of hydroxy-free radicle was tested by spectrophotometer. The expression of NF- κ B and Caspase-3 was investigated by Immuno-histochemistry technique.Results:1. Outcomes of neurological function grade: After recovering from anesthesia, all rats in MCAO saline control group appeared anergic in right anterior-extremity, prone to right spin, or to right side while walking or couldn't walk.
