Research Of Survivin Expression By Interference RNA In 5637 Bladder Cancer Cells

Objective To study the inhibitory effect of RNA interference (RNA i) on suvivin expression in bladder cancer cell line, 5637。Methods 5637 cclls were transfected with synthetic suvivin sequence-specitic siRNA by Lipofectamrne. The expression of suvivin mRNA was indenitied by quantitive PCR. Apoptosis of the transfected cells was examined by flow cyometry.Result the5637 cells was transfected dsRNA-survivin,the expression of surviving mRNA was suppressed markedly.Compared with control,the inhibitory rate of surviving mRNA was 29% and 53%,24 hours after transfection with the 40nmol/L specific siRNA,when the density of siRNA was 100noml/L,the inhibitory rate of surviving mRNAwas 46% and 86%.Apoptosis was identified in the identified in the transfected 5637 cells.Conclusion The expression of survivin at transcriptional levels in 5637 cells transfected with siRNA is markedly inhibited. And may be associated with the induction of apoptosis, That may be a new way to cure bladder cancer.
