For the purpose of studying the role of NGF (nerve growth factor, NGF) in the development of adenoid cystic carcinoma, we firstly investigated the possibility of culturing adenoid cystic carcinoma cell in serum-free medium, and based on this possibility, we studied the effects of NGF on the proliferation of in vitro cultured adenoid cystic carcinoma cell line Acc-2 and the lung metastasis cell line Acc-M by MTT assay. At last we used RT-PCR to detect the expression of NGF receptors TrkA and p75 in the cell lines on mRNA level.Part â…  Study of the growth of human adenoid cysticcarcinoma cell line in serum-free medium Objective: To observe the growth of human adenoid cystic carcinoma cell line (Acc-2) in serum-free medium.Methods: Acc-2 cell line was cultured in RPMI1640 containing ITS liquid supplement or 10% Bovine Calf Serum. The morphological changes were observed using phase-contrast microscope. Growth curves were made out under 3 circumstances: 1. Inoculating and culturing cells using serum-containing medium; 2. Inoculating and culturing cells using serum-free medium; 3. 24h after inoculating cells using serum-containing medium, cells were then cultured in serum-free medium. Rate of plating efficiency and cell adherent rate 24h after inoculating with density of 10~4/cm~2 were also compared respectively between cells cultured in the two mediums. At last, cell cycle analysis was made in cells cultured in both mediums by flow cytometry.
