Purpose: Dendritic cells (DCs) are extrely efficient antigen presenting ceils (APCs) which are potent stimulators of native T cell, even B cells to induce immune responses agasiot pathogens. Peripheric DCs have appeared to be central to immune system because of their ability to capture and process antigen, express costimulatory molecular and initiate a primary immune response. With discovery of diverse tumor-associated antigens and development of the methods for generation of DC and CTLs, current attempts such as cancer vaccines or adoptive transfer of T cells have been tested in clinical field.Tumor vaccines using dendritic cells have been shown to induce anti-tumor CTL responses.Based on the recognition of the central role of DCs in initiating immune responses, a variety of strategies have been devised to use DCs to stimulate immunity against tumor Ag's. Most of these strategies rely on the activation and maturation of DCs ex vivo and their subsequent reinfusion to tumor-bearing recipients after a pulse with tumor Ag's expressed.How to improve specificity of DC is now a new question. A number of previous studies already demonstrated trial that dendritic cells vaccine can induce specific immune response against and make tumor disappear in many animal tumor models .someclinical trials had been done in patients with melanoma, prostate cancer or renal cell carcinoma and clinical effects had been evaluated. Recent studies demonstrated the ability of DC to capture apoptotic cells and elicit MHC class I-restricted secondary CTL responses. Using DC pulsed with antigen or peptide can be successfully applied in tumor immunotherapy. This study aims induced culture DC in Vitro, through different types of leukemic cell antigen activation. Tumor vaccines using dendritic cells Its have been shown to induce anti-tumor CTL responses.
