The chemical compositions of Naematoloma fasciculare were studied systemically in this dissertation, and the anti-tumor effect of both fractions and compounds were examined using cell growth inhibition assay in vitro and the method of anti-tumor activity in vivo.The dry fruitbodies of N. fasciculare were extracted with MeOH 3 times at 60℃. The extract was evaporated and dissolved in water; then the suspension was defatted three times by petroleum ether then extracted with ethyl acetate. The water solution was evaporated and dissolved in MeOH. The sediment was dissolved in water again. Then the exact were departed into 4 parts, including the petroleum ether fraction, the EtOAc fraction, the MeOH fraction and the H2O fraction. The petroleum ether fraction was subjected on silica-gel column with a Cyclohexane:EtOAc (6:5) elution solvent and gave compound 1 and compound 4. The EtOAc fraction was concentrated and deposited for 24 hours; the white sediment was purified with diethyl ether and gave compound 2. And then the fraction of EtOAc solution was subjected on silica-gel column with a Cyclohexane:EtOAc (6:5) elution solvent and gave 16 fractions. Compound 4 was purified from D-3. D-7 was subjected on silica-gel column with a EtOAc (100 %)elution solvent and gave compound 3. And compound 5 was purified with EtOH from the water fraction. Compound 1-5 were identified as ergosterol peroxide, fasciculol C, cerevisterol, ergosterol and Mannitol using spectroscopic methods, including HPLC, NMR and MS and so on.The polysaccharide was extracted from N. fasciculare using the traditional method, then purified. The result indicated that the content of it was quite high, and can reach 3.6%.The petroleum ether fraction was analyzed by GC-MS, the chemical compositions were indentified into 20 compounds, induing 9,12-Octadecadienoic acid and palmitic acid and so on.The effects of the petroleum ether fraction, the EtOAc fraction, the MeOH fraction, the H2O fraction, crude polysaccharide and fasciculol C on cellular proliferation was proved with the MTT assay. The result indicated that the Petroleum ether fraction showed anti-proliferation activity compared with the 1640 medium-treated group in a concentration-dependent manner. The inhibition ratio of it was 60% at a concentration of 2mg/mL. In addition, fasciculol C showed anti-proliferation activity compared with the 1640 medium-treated group in a concentration-dependent manner, and then the inhibition ratio of it was 35.4%.
