Syringa pinnatifolia Hemsl .var. alashanensis Ma et S.Q.Zhou(shanchenxiang) belongs to the family of Oleaceae, genus of Syringa L., and its alias is Alashan Syringa. It has been used by people in Inner Mongolia for several hundred years as the substitute of Lignum Aquilariae Resinatum for the therapy of angina pectoris, heart-heat, dizziness, insomnia, cardiopalmus, asthma and"Heyi"disease. Satisfactory curative effect has been displaied by shanchenxiang. However, there are few researches on its chemical composition, pharmacological action and quality control. This situation is disadvantageous for the exploitation of shanchenxiang. In this thesis, the chemical composition of shanchenxiang was studied systematically. Then the protective activity of simple substances on hydrogen dioxide damage of neonate rat's cardiac myocytes was researched. Finally, a HPLC method for the simultaneous determination of two active lignans compounds was established, which could provided reference for the quality control of shanchenxiang.Twelve compounds were isolated from the ethyl ether fraction by column chromatography, gel column chromatography, flash chromatography, high speed countercurrent chromatography and high performance liquid chromatography. According to the NMR datas, seven compounds'structures were identified, which were 3- hydroxide-eremophilane-3, 11-diene-2,9-diketone(1), (-)-cubebin(2), lariciresinol(3), secoisolariciresinol(4), vanillic acid(5), 3',4'-acetal-lariciresinol(6) and (+)-dihydrocubebin(7). The compound 1 was a new compound that had never been reported before, and the other compounds were isolated from shanchenxiang for the first time.The activity research was performed using the oxidative damage in cardiac myocytes model to screen active compounds, which could protect the cardiac myocytes away from the oxidative damage by hydrogen peroxide. Lariciresinol and secoisolariciresinol exhibited extremely conspicuous protection activity. The protective effect of lariciresinol with a concentration of 10μg·mL-1 on cardiac myocytes was 13.5%, while the protective effect of 50μg·mL-1 was 29.3%. The protective effects of secoisolariciresinol were 16.7% and 31.1% respectively.In order to elevate the quality standard and strengthen the quality control of shanchenxiang, a HPLC method was established to determin the contents of lariciresinol and secoisolariciresinol simultaneously. A Kromasil 100-5 C18 column (250 mm×4.6 mm,5μm) was utilized. The mobile phase was methanol- acetonitrile-water(39:1:60) at a flow rate of 0.9 mL·min-1, and the jnject volume was 10μL. Through out the analysis, the temperature was kept at 35℃. Many experiments were carried out to validate this accurate method. Finally, the contents of lariciresinol and secoisolariciresinol from three shanchenxiang samples were determined by this HPLC method. This work provided experimental foundation for the development of the quality standard and quality control of shanchenxiang medical material.
