Obesity is commonly called rich man's disease. As the level of people's life are better off, obesity which not only affects figure and activity but also can cause hyperlipoidemia, artherosclerosis, coronary heart disease,hypertension,diabetes, arthrolithiasis, adiposis hepatica and so on,will be given more importance to it can be predicted that obesity will become one of mainly kinds of diseases that can threat our health.Obesity results from imbalance between energy intake and energy expenditure in which lipase and pancreatic lipase are critical. Fat tissue is hydrolyzed and released fatty acid as well triolein by lipase; on the other hand, high fat diet can't be directly absorbed across the intestinal mucosa.it must be firstly decomposed into one 2-monoglyceride and two free fatty acids by pancreatic lipase ,which is delivered into the lumen of the gut as a constituent of pancreatic juice. In other words, anti-obesity mechanism of drug lies in decomposing unnecessary fat tissue and inhibiting absorption of fat from food . exciting lipase or inhibiting pancreatic lipase can serve anti-obesity effect.Due to lower price of basal part of stem of notoginseng,what's more,it has higher content of ginsenosides, we attempted to group notoginseng saponins through silica gel dry column chromatography, then used column chromatography of ODS filling which just used as filling of analiytic column on HPLC to isolate monomer ginsenoside. Result has shown that the filling of ODS was more suitable to isolate ginsenosides from notoginseng saponins, and was more simple than filling of silica gel. In this experiment, we have isolated 5 ginsenosides ( i.e. ginsenoside Rb1, Rc, Rb2, Re, Rg1) which was identified by TLC, HPLC and MS spectra.on the other hand, we have found that many fators could affect the isolation of saponins such as particle size of filling which could directly affect the rate of eluent,amount of sample and eluent concentration which could affect the effectiveness of isolation.Ginseng saponins have three types: ①Protopanaxadiol—A type;②Protopanaxatriol —B type;③Oleanolic acid—C type; A and B types belong to four circles terpenoid ,but C type belongs to five circles terpenoid. Escins which have the same structure of C type of saponins belong to five circles terpenoid. On the basis of mechanism of anti-obesity, we have reserched whether saponins and Escins had effect of anti-obesity.Result has shown : American ginseng saponins and ginseng saponins had inhibitory action on pancreatic lipase in vitro, especially American ginseng saponins and ginsenoside Rb1,Rb2, Rc which belonged to panoxadiol have conspious inhibitory action ,and the inhibitory rate were...
