Objectives HBV infection is the serious worldwide public health problem.Each year approximately 50-120 million deaths occur as the consequence of HBV-related disease.It is well known that the chronic course of HBV infection is result of many factors,including virus , host, environmental factor and so on.The scientis want to find the genetic factor of HBV infection by investigating the functional genetic various site of the predisposing genes.we chose the T box expressed in T cells gene (TBX21)as the emphasis of the study.The aim of this study was to investigate the association between the single nucleotide polymorphisms (SNPS) of TBX21 Gene promoter and the susceptibility of hepatitis B in North Chinese.Methods Genomic DNA of 208 hepatitis B patients and 213 healthy controls were extracted from the peripheral blood leukocytes. Then two fragments covering position-1499 and position-1514 of the TBX21 Gene promoter were amplified by polymerase chain reaction (PCR). The SNPSs of patients was revealed by restriction fragment length polymorphism (RFLP), the control by DNA sequencing. We use the software PHASE 1.0 to construct the haplotypes of every individual. At last the unconditional Logistic regression model was used to analyze the statistical association of genotypes or haplotypes in two groups adjusted by gender and age.Results The two polymorphism positions of TBX21 Gene promoter possess the polymorphism in North Chinese (G → A on the position-1499, T→ C on the position-1514) . Compared Genotypic frenquency between G/A and G/G of the polymorphism on the position-1499,the difference is obviously P<0.05 OR=1.76 (95%CI: 1.03-2.99).Otherwise the difference is significant in the comparison of Genotypic frenquency between A/A and G/G.
