Studies On The Chemical Components In Kidney-tonifying And Yang-boosting Part Of Curculigo Orchioides Gaertn

Rhizoma curculiginis is the dried rhizomes of Curculigo orchioides Gaertn. As a commonly used Sichuan's medicinal substance, it is also recorded in Pharmacopoeia of China. Rhizoma curculiginis is bitter in taste and warm in nature, Its actions include kidney-tonifying and yang-boosting, strengthening the muscles and bones, expelling wind and rheumatism, etc. On clinic's application, it has been used to treat impotence, lumbago, cold diarrhea, etc.Curculigoside is a kind of phenolic glycoside. As an active component and standard sample, it has been shown to enhance phagocytic activity of macrophages, stimulate immune response by acting both on macrophages and lymphocytes. But its pharmacological activity has no definite relation to kidney-tonifying and yang-boosting. So kidney-tonifying and yang-boosting part of Curculigo orchioides Gaertn was screened on castrated mice. The results indicate that the kidney-tonifying and yang-boosting part is gained by extraction using 80% ethanol and following abstraction with n-butanol. The extracts were chromatographed on the Macroporous absorbent resin column and repeatedly chromatographed on the silica gel (SiO₂) column using gradient elution process. Then, it was further purified by recrystallization using various organic solvents. Four compounds were finally isolated. The structures of these compounds were identified by means of NMR, MS, etc. Their structures were identified as curculigoside, curculigoside B, curculigine A, 2,6-dimethoxy benzoic acid. Two components (curculigoside and curculigine A) with higher content were studied by pharmacodynamic experiments. Curculigine A has certain activity. It was detected by HPLC.
