Object:To investigate the effects of salvia miltiorrhiza and its essential components on tumor growth and metastasis and make an preliminary study of the involved biological mechanisms. Method: (1) In vivo experiment of tumor inhibitition . Models of tumor growth in mice with S180 sarcomas were used to observe the differences of their general states and ten-day neoplastic weight between control group and Danshen treatment group. Observe the morphologic changes of tumor cells by HE staining and test the expression of VEGF,ICAM-1 and VCAM-1 by the immunohistochemical method. (2) Test the effects of Salvia miltiorrhiza and its essential components on the proliferation of highly metastatic lung carcinoma cell lines(PG) and human jugular vein endothelial cell lines(HJVEC) by MTT colorimetric method in vitro. (3) Label the PG tumor cells with fluorescence , and observe the effects of Salvia miltiorrhiza and its essential components on the adhesion between PG cells and HJVEC cells. Results : The tumor in salvia miltiorrhiza group weighed significantly less than that in the control , but the expression of VEGF displays no significance compared with the control, which indicates that how Salvia miltiorrhiza inhibits tumor growth was irrelevant to the low level of expression of VEGF. In contrast to the control , the expression of ICAM-1 and VCAM-1 in tumor tissues significantly diminished , which possibly suggested the blood circulation promotion of salvia miltiorrhiza . The experiments in vitro showed that Sal B was able to promote the proliferation of HJVEC cells, however , it had no effect to inhibit the tumor growth as well as the adhesion between PG cells'and HJVEC cells. Tanâ…¡A could inhibit the proliferation of PG cells and HJVEC cells and decrease the adhesion between PG cells and HJVEC cells. Compound injection of Salvia miltiorrhiza could inhibit the proliferation of tumor cells but strengthen the adhesion between PG cells and HJVEC cells.
