Objective: To observe the effects of Crocin on human bladder cancer T24 cell line and to explore the anti-cancer mechanism.Methods:â‘ T24 cells for human bladder carcinoma were cultured by cell culture technique. The MTT assay was used to evaluate the inhibitory effect of Crocin for growth of T24 cells.Microscope was used to observe the morphological changes of the cells. Flow cytometry was used to measure the cell cycle and cell apoptosis.RT-PCR and Westernblot were used to detect Bcl-2 and Bax expressions.â‘¡T24 cells were inoculated into the right-back legs of BALB/c nude mice to establish bladder cancer model. After treatment with Crocin, the inhibitory effects were observed on the growth of tumor. Microscope and electron microscope were used to observe the morphological changes. RT-PCR was used to detect Bcl-2 and Bax mRNA expressions. The expressins of Bcl-2,Bax,survivin and CyclinD1 were detected by immunohistochemistry.Results:â‘ In vitro: The growth of T24 cells was inhibited remarkably in the way of dosage dependence and time dependence. After the T24 cells were treated by Crocin, distinct morphology changes of cell apoptosis were observed by Giemsa staining. Flow cytometric profiles revealed that Crocin led to the increase of the cell percentage of G0/G1 phase and the cell percentage of apoptosis.
