Objective: Dendritic cells is the dominant antigen-presenting cell of the immune system, and play an key role in cancer immunotherapy. Gastric cancer cells are lack well-characterized tumour-specific antigens and low immunogenic, the former methods: gastric carcinoma antigen pulsed DCs and cancer DNA,RNA transfect DCs ,which irritated DCs to bring about tenuous anti-tumor immune reactions. In this experiment, cell fusion technology was used to get fusion vaccine of the gastric cancer cells and mature dendritic cells which were generated from human umbilical cord blood in vitro, then the biological characteristics and the efficacy of antitumor function about allogeneic DCs-gastric cancer cell fusion vaccine was studied in vitro. we hoped to establish a foundation and provide theoretical guidance for further study in vivo and clinical application.Methods: First, mononuclear cells were separated from human umbilical cord blood , mature dendritic cells were generated depending on co-cultured with rhGM-CSF,rhIL-4,TNF-a. The allogeneic DCs and SGC7901 cells were fusioned by using of 50% PEG and the pure FCs were screened out by adherent incubate.The morphology of DCs,SGC-7901 and FCs was respectively observed by light,electron microscope testing and staining method.
