For the purpose of systematic quality evaluation of Chrysanthemum morifolium Ramat. and its extraction method, the following studies were carried out.1. the standard substance of 3,5-O-dicoffeoylqunic acid,luteolin-7-O-β-D-glucoside, luteolin, acacetin-7-O-β-D-glucoside were isolated from Chrysanthemum morifolium Ramat.and the structure were elucidated by UV, IR, MS, 1H-NMR and 13C-NMR. The purity of 4 standards substance were over 98% by HPLC.2.according to literature's recordation, Mophology of different sources of Chrysanthemum morifolium Ramat.were carried out.3. Reversed-phase high performance liquid chromatography(Rp-HPLC) method wasused for the assay of 2 caffeoylqunic acids compounds (chlorogenic acid,3,5-O-discaffeoylquinc acid) and 4 flavonoids (luteolin-7-O- β -D-glucoside, apigenin-7-O- β-D-glucoside, luteolin and acacetin-7-O- β-D-glucoside). The gradient solventsystem(acetonitrile-water-acetic acid) and ODS reversed-phase column of Hypersil C18(4.6 mm×250mm, 5μm) were used in the quantitative analysis of the 6 components.The eluate was monitored with UV absorption at 348nm. The linear ranges of chlorogenicacid, luteolin-7-O- β-D-glucoside, 3 , 5-O-discaffeoylquinc acid, apigenin-7-O-β-D-glucoside, luteolin and tilianin were in 0.006~0.117 mg·mL-1, 0.002~0.046mg·mL-1,0.052~1.030 mg·mL-1,0.010~0.202 mg·mL-1,0.002~0.050 mg·mL-1 and 0.003~0.052mg·mL-1 (r≥0.9995) respectively. The recoveries were in 97.2~100. 7%, and the RSD wereless than 2.5%. The methods are simple, rapid and with good reproducibility and provide aquantitative basis for the quality assessment for Chrysanthemum morifolium Ramat.
