| Objective: A preliminary study of the pharmacodynamics of otal flavonoids by using avariety of methods. Compared the pharmacokinetics and myocardial ischemia of totalflavonoids (TF) and nanoemulsion (TF-NE).Methods: DPPH method for the determination TF of anti-free radicals; intraperitonealinjection of STZ diabetic rats were prepared the observed total flavonoids diabetic ratsclotting time in vitro thrombus length and dry, wet weight, whole blood and plasmaviscosity., hematocrit and erythrocyte sedimentation rate, and serum nitric oxide (NO)content. The HPLC gradient elution, the simultaneous determination of TotalFlavonoids of three flavonoid aglycones; prepared total flavonoids nano-preparation,the establishment of a method for determination of rat processing of biological samplesand measured, and TF, TF-NE pharmacokinetic study. Coronary ligation to induceischemia model, determination of serum enzymes (AST, LDH, CK) activity, clottingtime, and infarction rates compare total flavonoids nanoemulsion difference.Results: TF have strong anti-DPPH radical role; total flavonoids can significantlyprolong the the diabetic rats clotting time, reduce the length of the in vitro thrombosisand dry, wet weight, lower hematocrit, slow down the erythrocyte sedimentation rate,elevatedserum NO content; realized by RP-HPLC Determination of total flavonoids ofluteolin, apigenin and three kinds Acacetin, flavonoid aglycones; TF-NE werecharacterized by the average diameter of83.16±12.03nm, the zeta potential of-(11.62±6.38) mV, relatively narrow particle size, the lower the system stability; TF-NEmorphology observed by transmission electron microscopy (TEM), formulated in aspherical or spherical, more uniform particle size distribution, no adhesion. Rats wereTF-NE compared with the TF-plasma luteolin and apigenin Tmaxhas pushed theabsorption half-life and elimination half-life of a certain degree of extension of a certainincrease in Cmax, TF-NE in the luteolin AUC is2.52times the bulk drugs, thenanoemulsion apigenin in the AUC is3.62times the bulk drugs; luteolin and apigenin relative bioavailability of313%and318%, respectively. Compared with model group,orally TF and TF-NE rat serum enzymes (AST, LDH, CK) activity has decreasedcoagulation time infarction rate also decreased, but no significant difference betweenTF and TF-NE.Conclusion: TF can anti-free radical, anti-oxidation, improve blood rheologyabnormalities. Nanoemulsion improves the the bioavailability of total flavonoids, TFand TF-NE has against acute myocardial ischemia. |
