Objectives: To aim at femoral veins gained from the rats with acute traumatic limbs deep vein thrombosis (DVT), through Affymetrix microarray technique, the genes related to fibrolysis-antifibrolysis system in 6 crucial phases of thrombi formation and variation were studied further. From the genes appearing significantly differential expression in fibrolysis-antifibrolysis system, tissue-type plasminogen activator (t-PA) and plasminogen activator inhibitor-1 (PAI-1) were selected and detected through the fluorescent quantitation real time RT-PCR technique in order to provide new experimental data for clinical diagnosis and therapy of DVT.Methods: 1. The experimental animals were divided randomly into 7 groups: control group (group A), traumatic instant group (group B), prethrombotic group (group C) thrombosis crest-time group (group D), thrombi solution group (group E), thrombi insolution group (group F), non-thrombosis group (group G). Total mRNAs of femoral vein specimens from the 7 groups were extracted separately. Then microarray detection was performed according to the manipulation process of Affymetrix RAT 230A microarray. All data were dealt by Microarray Suite5.0 software so as to initially analyze and summarize the expressions of genes in blood coagulation, fibrolysis, inflammation and immune systems.2. To aim at fibrolysis and antifibrolysis system, the related genes were selected from the databases of National Center for Biotechnology Information (NCBI) and China National Knowledge Infrastructure (CNKI). Combined with the data gained from Affymetrix RAT 230A, the Rat Genome U34 Set in Unigene database(build 34, GeneBank107/ dbEST11/18/98, GeneBankl10 supplement) was used to build a Cluster tree in order to analysis the expressions of genes related to fibrolysis-antifibrolysis system.
