Objective: To explore the change in expression of nerve growth inhibitor Sema3A in spinal cord and its receptor neuropilin-l(NP-l) in spinal cord and spared dorsal root ganglion after unilateral partial dorsal root rhizotomy of adult cats. Furthermore, to offer datas for exploring roles of nerve growth inhibitor in spinal cord plasticity.Method: 15 adult cats were used. They were divided into 3 groups: normal control group, unilateral partial dorsal root rhizotomy 7th and 14th day groups ( i.e. rhizotomy of unilateral L1-L5, L7-S2 dorsal roots, leaving L6 dorsal root as a spared root . The animals were allowed to survive 7 days and 14 days). The animals were purfused and fixed with 4% paraformaldehyde .Their L3, L5, L6 segment of spinal cord were made into 35 μm thickness frozen sections and L6 dorsal root ganglia (DRG) in operated side were made into 20 μm thickness sections. Sections of spinal cord were stained with specific Sema3A (1:1600) and NP-1 (1:500) antibody by immunohistochemistry ABC method and L6 DRG were stained with NP-1 antibody (1:600). Mean optical density (OD) of Sema3A and NP-1 immunoreactivity in superficial laminae of the dorsal horn was measured with image analysis system. The number of...
