Objective: Although some studies found the association between glucocorticoid receptor gene N363S polymorphism and MS (metabolic syndrome)in Caueasians ,there is no report in Chinese.The purpose of this study is to compare the difference of genetic backgrounds among a metabolic syndrome(MS) group,group with glucose ,lipoids metabolic disorder,but not enough to diagnostic criteria of MS,and normal healthy controls, and to explore whether glucocorticoid receptor gene N363S polymorphism are one of the genetic disorders which could cause the MS in Chinese,expected to find new mutable site.Methods: the 357-bp area of exon 2 of the glucocorticoid receptor gene was screened by polymerase chain reaction-single strand conformation polymorphism analysis(PCR-SSCP)in all groups.DNA fragments displaying an abnormal migration pattern and some samples with normal migration pattern during SSCP analysis were subjected to direct sequencing. Results: Val271 Ala mistake sense mutation was detected from 1 subject in healthy control group. Pro274Gln mistake sense mutation was detected from 1 subject in healthy control group.
