Cornus is the ripe fruit of cornel that belongs to cornel genus,cornel family.lt grows mainly in the upland of the north temperate zone and semitropical zone. According to ben cao gang mu, a legendary medical book in China, cornus can strengthen jism and semen, and it smells vinegary and slipslop, and it has no toxicity to the body of people. Cornus has an effective cure for the chills and fever in the heart,and it can Warm the Middle-jiao and lustrate the cold and humidity in body,and eating it can strengthen the body.The modern medical research proves that cornus can strengthen organism immune ability, restrain inflammation, lower blood sugar, resist the exsanguine shock, and it can suppress blood platelet coagulation and has the antibacterial, antioxidant, anticancer effect.The total saponin is one main active element of cornus, and large researchs show that saponin has the biological effect of dispelling sputum, relieving a cough, restraining bacterium, resisting cancer, reducing fever and so on. So it is very important to research the extraction and purification technology and the biological effect of the cornus total saponin.The test uses the cornus as material.The optimal parameter of extracting the cornus total saponin by water, alcohol solvent and ultrasonic,the purification technology by HPD-100 resin, the antioxidant activity in vitro of the cornus total saponin have been studied in this paper. The result shows:1. The optimal parameters of the total saponin extraction from the cornus by water are as following: material/solvent ratio 1:12, extracting 3h at 70℃, for one time.2. The optimal parameters of the total saponin extraction from the cornus by alcohol solvent are as following: the concentration of ethanol 60%, material/solvent ratio 1:20, extracting 1h at 70℃, for one time.3. The optimal parameters of the total saponin extraction from the cornus by ultrasonic are as following: ultrasonic power 250W, the concentration of alcohol 60%, material/solvent ratio 1:16, extracting 20min.4. HPD-100 macroporous resin shows better performance for purifying the cornus total saponin, and its static adsorption ratio is 113.75mg/g dried resin, the elution ratio is 92.10%;The parameters of dynamic adsorption are as fowllowing: the original concentration 7~9mg/mL, the feeding rate 2BV/h and pH value 6.0 ~ 7.0;Theparameters of dynamic elution are as fowllowing: firstly eluting the water-soluble impurity like amylose by distilled water, then eluting by 50% alcohol, collecting the part eluted by 50% alcohol;the eluting cubage of 50% alcohol 250mL, the eluting rate l~2BV/h;the total saponin in the solid after purification is more than that in the solid before purification, the refinement ratio can reach 242.9%, which shows the effect that HPD-100 macroporous resin purifies the cornus total saponin is well.5. The scavenging capability of the cornus total saponin to superoxide anion radical is well, and is better than Vc. The scavenging capability increases with the concentration of the cornus total saponin rising, and the maximal scavenging ratio reaches 86.5%.6. The cornus total saponin and Vc both have good scavenging abilities to the hydroxyl free radical. With the concentration of the cornus total saponin rising, the ability clearing the hydroxyl free radical increases. The IC50 value of the cornus total saponin to clear the hydroxyl free radical is around 0.2mg/mL and that of Vc is about 0.8mg/mL, which indicates the cornus total saponin has stronger scavenging ability than Vc.7. For the cornus total saponin and Vc, the scavenging capabilities to alkane radical both increase with the concentration of the cornus total saponin and Vc rising. The IC50 value of Vc and the cornus total saponin to clear alkane radical are 0.8mg/mL and 1.6mg/mL, and the maximal scavenging ratio to alkane radical of Vc is 68.7% and that of the cornus total saponin is just 52.8%, which indicates the cornus total saponin and Vc both have quite scavenging capabilities to alkane radical and the scavenging ability of the cornus total saponin is stronger than that of Vc.8. The cornus total saponin and Vc both have obvious scavenging capabilities to DPPH radical. In DPPH system, the IC50 value of the cornus total saponin and Vc are 0.2mg/mL and O.lmg/mL, but the scavenging capability of the cornus total saponin is parallel to Vc in the high concentration. In total, the scavenging capability to DPPH radical of the cornus total saponin is weaker than Vc.
