Object: The study was to observe the protective effect of taurine on apoptosis and the production of NO induced by ox-LDL in cultured vascular endothelial cells(ECs).Methods: Cultured human umbilical vein endothelial cells (ECV304) were used for the experiment to investigate the protective effect of taurine against the damage of ECs induced by ox-LDL. This study were divided two parts: The first one was to explore the damage of ECs induced by ox-LDL. Four groups were divided: the control group, 25mg/L ox-LDL group, 50mg/L ox-LD group and 100mg/L ox-LDL group; The second part was to investigate the protective effect of taurine on the damage caused by ox-LDL. six groups were divided: the control group, ox-LDL(50mg/L) group, Taul (taurine 5mmol/L + ox-LDL50mg/L) group , Tau2 (taurine 10mmol/L + ox-LDL50mg/L) group, Tau3 (taurine 20mmol/L + ox-LDL50mg/L) group and Tau4(taurine 40mmol/L + ox-LDL50mg/L) group. Cytoactive were detected by the method of MTT, cell apoptosis were determined by TUNNEL and flow cytometry(FCM), the production of NO was measured with nitrate reduction test.Result: The first part: The effect of ox-LDL on the cytoactive and apoptosis in ECs1. Incubation of ECs with ox-LDL for 24h , the cytoactive decreased in a concentration-dependent manner. Comparing with the control group( 1.00+0.07), the cytoactive of 25mg/L,50mg/L and 100mg/L ox-LDL group were 81% (0.81±0.16 P<0.05X 72% (0.73 + 0.08 P<0.01), 59% (0.59+0.17 P<0.01) respectively.2. After ECs were Exposed to ox-LDL for 24h ,the rates of apoptosis increased in a concentration-dependent manner. Comparing with the control group (1.37+0.15%) , the rates of apoptosis of 25mg/L,50mg/L and 100mg/L ox-LDL group were 11.7-fold (16.25 + 2.28% P<0..01), 16.9-fold (23.53+3.77% P0<.01),22.3-fold (31.20 + 3.21% P<0.01) respectively.The second part: The protective effect of taurine on the cell apoptosis and...
