Objective This study was undertaken to evaluate associations of the -675 4G/5G PAI-1 polymorphism with plasma PAI-1 in asthmatic patients, and to explore the possible roles of -675 4G/5G PAI-1 polymorphism in asthmatic remodeling. Methods Genomic DNA was isolated by the salt fractionation method from the peripheral blood of 99 asthmatic patients and 101 healthy subjects. The PAI-1 4G/5G genotypes were determined by an allele-specific polymerase chain reaction. 30 chronic persistent asthmatics of 99 asthmatic patients after 3~6-day treatment and 20 control subjects performed spirometry and HRCT to for the evaluation of the severity of asthma and airway remodeling. Quantitative assessment was used to analyze the thickness of airway wall (T, T/D) and airway wall area (WA, WA%). Levels of PAI-1, TGF-β1 and MMP-9 in plasma were detected by ELISA.Result 1.The 4G/5G PAI-1 polymorphism in asthmatic patients: (1) Genotype distributions were in Hardy-Weinberg equilibrium, both within each group and in the population as a whole. Frequencies of genotypes in asthmatics [4G/4G (0.374), 4G/5G(0.495), 5G/5G (0.130)] differed significantly from those of controls [4G/4G(0.277),...
