Evodia rutaecarpa., a traditional Chinese drug, which is the fruit of Evodia rutaecarpa. (Juss) Benth, Evodia rutaecarpa. (Juss) Benth. var. officinalis( Dode) Huang, Evodia rutaecarpa. (Juss) Benth var. bodinieri (Dode) Huang.The chemical constituents of Evodia rutaecarpa are complex, so its drug efficacy is various . But the study on its antitumor activity is few.For studying and developing Evodia rutaecarpa. better, further studying the Evodia rutaecarpa. effective component, determining its antitumor active spot, we have carried through the studies of systemic chemical constituents from Evodia rutaecarpa.. And some of the obtained compounds were tested on the antitumor activity. Taking the active test result as the instruction, we optimize the lead compounds' structure to obtain the high-activity compounds.In the part of extracting and isolating active compounds, the alcohol was used as extracting solvent. Then the extraction was parted in different fraction using different polar solvent as petroleun ether, ethyl acetate, n-butyl alcohol, and methanol. 17 coumpounts were isolated altogether. 12 compounts structure were determined on spectral evidences and physics and chemistry constant as rutaecarpine (WZY-1) , evodiamine (WZY-2), 14-formyldihydrorutaecarpine (WZY-3), 1-methy-2-[(6Z, 9Z) -6,9- pentadecadienyl ] - 4(1H) -quinolone (WZY-4), eicosanoic acid butyl ester (WZY-5), lignoceric acid (WZY-6), β-sitosterol (WZY-7), palmitic acid (WZY-8), dihydroevocarpine (WZY-9), 1-methyl-2-(11-tetradecenyl)-4(1H )-quinolone (WZY-10), β-daucosterol (WZY-11), evocarpine (WZY-12). Compounds eicosanoic acid butyl ester (WZY-5), lignoceric acid (WZY-6) and 1-methyl-2-(11-tetradecenyl)-4(1H )-quinolone (WZY-10) were obtained from this plant for the frist time. In this article, the polysaccharide of Evodia rutaecarpa. was also extracted and suprified.The antitumor ativity of 5 alkaloids and polysaccharide from Evodia rutaecarpa. were studied by using MTT assays to measure the inhibitory effect to K562 cell line, and SRB assays to measure the inhibitory effect to A549 cell line respectively.
