PURPOSE: To test the expression of PCNA and MMP-2 in the operating area.To determine whether application of a broad-spectrum matrix metalloproteinase (MMP) inhibitor, GM6001, reduce scarring after glaucoma filtering surgery.METHODS: 24 New Zealand white rabbits were randomly divided into four groups. Each group had 12 rabbits. We operated on every rabbit's eye according to standard trabeculectomy.3 groups received GM6001 cotton swab of 3000μM, 2000μM and 1000μM for 5 minutes, respectively,1 group received nothing during the operation. The animals were killed on days 32. We observed the rabbits' filtering blebs, corneal epithelium and conjunctival wound on postoperative days and Intraocular pressure was recorded. We observed the filtering passages by performing HE staining. Immunohistochemistry analysis were used to determine the expression and changes of PCNA and MMP-2.The numbers of PCNA positive cells were counted.The datas were tested by statistical analysis.RESULTS:1.After surgery ,the Intraocular pressure (IOP) of all groups droped.The IOP was significantly lower in the treated groups (GM6001) than in the control group(P<0.05).2.The surviving time of the functional blebs were showed statistically significant differences between the control and treated groups (P<0.05).
