Activation of certain ongene and deactivation of specific anti-oncogene are concernd in the occurrence and development of different kinds of tumors. The family of Runx gene consists of Runx 1, Runx2 and Runx3. Runx3 gene is located in the zone of 1p36.1 in human chromosome and recently recognized as anti-oncogenic, which is potentially a key gene inducing the occurrence of gastric carcinoma. It is repeated reported in a variety of literature that Runx3 gene is intimately correlated with gastric carcinoma through frequent methylation of the promotor zone. The correlation is 10-fold higher than any other known anti-oncogene has.Method: 40 gastric cancer samples were collected from the department of general surgery of the fist and second facilitated hospital of Jilin University, and were identified by pathology. The expression of runx3 mRNA in the samples and normal gastric membrane was detected using the reverse transcription- polymerase chain reaction. Then analysis was carried out to find the correlation between the expression and certain clinical parameters.Result: The relative expression value of runx3 in gastric cancer tissue (0.5452±0.1285) was lower than that of the control group (0.9526±0.0532).The expression difference between the two groups was significant statistically (t=4.891, P < 0. 05).The same results could be harvested by comparing the values between the undifferentiated or poorly differentiated(0.4867±0.0758) and moderately with or highly differentiated gastric carcinoma groups(0. 7991±0.1945) with significance of (t=4.796,P<0.05), and between the metastatic group(0.5090±0.1263) and the group with no lymph node metastasis(0.7823±0.1502)with significance of (t=4.206,P <0.05). Gender and tumor size weren't significantly correlated with the expression of runx3 runx3 mRNA.Conclusion:1.The expression of runx3 mRNA in gastric carcinoma tissue was lower than that in the control group with statistical significance, and was correlated with the pathologic type and metastasis.2. The expression of runx3 gene plays an important role in the genesis and development of gastric cancer.
