Objective: To investigate the expression of maspin ,uPA and MMP-7 in gastric cancer and the relationship between clnical pathological parameters. To explore their roles in tumorigenesis and progression of gastric cancer.Materials and methods:Immunohistochemistry SP method was used to detemine expression of maspin, uPA and MMP-7 in gastric adenocarcinoma(n=30), signet-ring cell carcinoma.(n=30), normal gastric mucosa (n=20). The reverse transcriptase PCR was carried out to detect the level of maspin mRNA from the above mentioned 30 gastric adenocarcinoma and 20 normal samples.Results:Inmmunohistochemistry:(1)Of 30 cases of gastric adenocarcinoma, the positive rates of maspin,uPA, MMP-7 were 50%,70%, 80%, respectively; Of 30 cases of signet-ring cell carcinoma , the positive rates of maspin,uPA, MMP-7 were 46.7%,76.7%,90%, respectively.Of 20 cases of normal gastric mucosa, the positive rates of maspin, uPA, MMP-7 were 90%, 35%, 30% ,respectively. (2)Maspin expression significantly related to invasive depth, metastasis, but not to tumor size and TNM staging. The expression of uPA and MMP-7 significantly related to invasive depth, metastasis and TNM staging, but not to tumor size.(3)maspin expression had a positive correlation with uPA and MMP-7, uPA expression had a positive correlation with MMP-7. RT- PCR : The relative level of maspin...
