PART I Effect and mechanism of HXMTZG on the diabetic nepropathy rats1. Effect of HXMTZG on weight, drinking, eating and blood glucose in diabetic nepropathy ratsObjective: To study the influence of HXMTZG on weight, drinking, eating and blood glucose in diabetic nepropathy rats. Method: In regular dieting rats are fed with the solvent of 30mg.kg-1 ip. STZ on the first day and on the next day with the solvent of 0.11 ml/one rat ip. FCA in a week, and it goes in this way for three weeks. Then the blood glucose of the rats is tested, and those whose blood glucose is≥16.7mM are divided randomly into four groups: control, aminoguanidine (0.1g.kg-1 , ig.), HXMTZG (0.25, 0.5g.kg-1, ig.),and the normal group is added. The rats are observed 12 weeks for resulting diabetic nepropathy. We measure the rats' weight and blood glucose every three weeks, drinking and eating once a week in order to track the dynamic changes of diabetic rats. Result: HXMTZG can remarkably reduce drinking and eating , increase weight but indistinctively and can't lower blood glucose in diabetic rats. Conclusion: HXMTZG is able to mitigate the symptoms of diabetic nepropathy rats at a certain extent.2. Protection of HXMTZG on kidney and other tissues in diabetic nepropathy ratsObjective: To investigate whether HXMTZG can protect kidney and other tissues (including thoracic aorta, heart and sciatic nerve)in diabetic nepropathy rats. Method: In regular dieting rats are fed with the solvent of 30mg.kg-1 ip. STZ on the first day and on the next day with the solvent of 0.11 ml/one rat ip. FCA in a week, and it goes in this way for three weeks. Then the blood glucose of the rats is tested, and those whose blood glucose is≥16.7mM are divided randomly into four groups: control, aminoguanidine (0.1g.kg-1, ig.), HXMTZG (0.25, 0.5g.kg-1, ig.) ,and the normal group is added. The rats are observed 12 weeks for resulting diabetic nepropathy. Then we adopt light microscope to observe the morphology of pathological damage of kidney, thoracic aorta, heart, and sciatic nerve. Result: After 12 weeks duration, every samples of the control show apparent pathological changes as follows: glomerulus divided to many branches, mesangium expanded, increased and eosin stained matrix; lipid vacuoles and clearing cast increased in epithelial cells of renal tubule, partly coagulating necrosis of renal tubule in medulla, inflammation cells shown in stroma and some developed to pyelitis. Bubble cells are found in the blood vessel wall of thoracic aorta and the blood vessel wall thicken at the place of bubble cells aggregation. As to the heart, there are many necrosis in myocardial cells and lots of neutrophil infiltrated in the necrosis place. The complication of the axon and myelin sheath in sciatic nerve is not significant. But all the pathological changes above are ameliorated by HXMTZG, which suggests that the component can protect kidney and other tissues from damaged by the diabetic nepropathy state.
