Objective: To explore the preconditioning effects and mechanism of SF on adult cultured myocardial cell injury induced by anoxia reoxygenation. Methods: Cultured myocardial cells of adult Wistar rats have been randomly divided into six groups: Control group: reoxygenation of 70min; anoxia reoxygenation group(A/R): reoxygenation of 30min following anoxia of 40min; anoxia preconditioning group(APC): reoxygenation of 30min following anoxia of 10min before the same procedure as A/R group; SF+A/R group: 20min of SF(0.42mmol/L) preconditioning followed by 20min wash out before A/R; Glib+APC+A/R group: adding Gliberclamide of 0.1 mM final concentration to cultured medium 30min before the same procedures as APC preconditioning group;Glib+SF+A/R group: adding Gliberclamide of 0.1 mM final concentration to cultured medium 30min before the same procedures as SF preconditioning group. At the end of experiment, myocardial cell viability and the activity of LDH in culture were measured, furthermore, The morphology or ultrastructure assessment of ventricular myocytes was observed by inverted microscope or transmission electron microscope. Results: ①the cell viability: The cell viability of A/R group(41.5±1.9%) was significantly decreased when compared with that of control group (p<0.01) ;The viability of APC+A/R group (74.5±3.3%) , SF+A/R group (73.5±3%) was significantly increased when compared with A/R respectively(p<0.01) , the cell viability was no significant difference between APC +A/R group and SF+A/R group (p>0.05) ; The viability of Glibenclamide+APC+A/R group(42.9±2.8%) and Glibenclamide+SF+A/R group was significantly decreased when compared with that of APC group (p<0.01) , while the cell viability was no significant difference between Glibenclamide+APC+A/R group...
