| Piper laetispicum C.DC., which only grows in China, belongs to the genus Piper, Piperaceae, widely distributed in Guangxi, Guangdong, Hainan provinces and some islands near the oceans. It is mainly used for injuries from falls, releasing ecchymosis and eliminating bump in folk medicines. Our group screened fifteen species of the genus Piper in South China, and found that the ethyl acetate fraction of the ethanol extract of Piper laetispicum C.DC. root and stem showed obvious activity against depression, moreover, we found a novel compound (designated laetispicine),it had evident activity against depression(Note: indicated by exp., the activity against depression of laetispicine is five times of Fluoxetine). There are more than thirty compounds have been separated and identified from the Piper laetispicum C.DC.. From the ethyl acetate fraction of the ethanol extract of Piper laetispicum C.DC. root and stem, eight compounds were separated and identified by Wang Ezhou, and fourteen compounds were separated and identified by Qu Gonglin. From the ethanol extract of Piper laetispicum C.DC. leaves, sixteen compounds were separated and identified by Jin Tao. Among the total, 5'-methoxy-3',4'-methylenedioxy-2Z-cinnamicacid isobutylamide is a new compound, separated and identified by Qu Gonglin.Twelve compounds were separated and purified by the means of silica gel, Sephadex LH-20, HPLC from the ethyl acetate fraction of the ethanol extract of Piper laetispicum C.DC. root and stem. Their structures were identified on the basis of UV,IR,MS,NMR and compared with the standard substances.As a result, compound I and compound II are novel ones, designated laetispiamide A and B. laetispiamide A show to be N-isobutyl-(3,4-methylendioxyphenyl)-2E,4E,7E-Nonatrienamide, laetispiamide B show to be N-isobutyl-(3,4-methylendioxyphenyl)-2E,7E-Nonadienamide.And compound III was separated first from Piper laetispicum CDC, it shows to be N-isobutyl-... |
