Objectives To observe the effects of tamoxifen on the growth of transplanted human hepatocellular carcinoma (HCC) in nude mice and the changes of estrogen receptor (ER) and transforming growth factor β1 (TGF- β1) indicators in carcinoma tissue of nude mice after filling tamoxifen suspension into stomach by creating models of transplanted tumor with HepG2 in nude mice; To study the effects and mechanism of tamoxifen on HCC; To explore new methods and provide theoretical and experimental evidence for supplementary treatment and postoperative chemotherapy with anti-hepatoma chemicals.Methods 26 BALB/cA-nu male nude mice were transplanted with 1.5 × 106 HepG2 human hepatocellular carcinoma line hypodermically on the back to create models of transplanted tumor. The growth of tumor was observed everyday. One nude mouse developing ascites on the 8th day because of abdominal metastasis and another one having extensive integumentary infiltration on the back 10th day were taken out. 22 nude mice developed tumors (84.6%) on the back in 14 days (delitescence: 6-12 days). All the 22 nude mice were randomly divided into experimental group and control group and weighed. The longest (a) and shortest length (b) of tumors were measured and used to calculate the tumor volume by the formula V = a×b2×0.52. The difference between two groups was compared. After filling tamoxifen suspension 4 mg/kg/day into stomach to experimental group and normal saline 4 mg/kg/day to control group for 30 days continuously all nude mice were killed by cutting neck. The tumors were removed, measured and weighed. The weight and size of tumors before and after experiment were compared in two groups and the inhibiting rate on tumor of tamoxifen was calculated.
