Objective:To explore the gemcitabine (GEM) for in vitro culture people subcutaneous hepatocellular carcinoma HepG2cells and nude mice transplantation tumor growth inhibition and its mechanism.Methods:HepG2in vitro human liver cells, were randomly divided into control group and gemcitabine group two groups.According to the GEM will join different concentrations in human hepatocellular carcinoma HepG2cells culture medium after48h, Were observed under inverted microscope cell growth condition, the cell growth inhibition rate determined by MTT method and flow cytometry assay, apoptosis percentage.Choose one hepatocellular carcinoma HepG2cells in nude mice subcutaneous transplantation tumor model, two groups were randomly divided into blank control group and the GEM group.After being a tumor-burdened nude mice grouping processing20d, observation of nude mice subcutaneously transplanted tumor tumor volume and draw the growth curve, immunohistochemical detection of tumors in the Bel-2and WWOX gene expression-Results:observed under inverted microscope:the GEM group a number of suspension cell death, cell growth and control group did not see abnormalities.Fluid cytology test results showed that different concentrations of1,25(2)5,5,10,20,40mg/L GEM role human hepatocellular carcinoma HepG2cells after48h, can promote human hepatocellular carcinoma HepG2cells apoptosis, and cell apoptosis percentage increase with the increase of its concentration, the dose-response relationship, significantly higher than the control group (P<0.05).Determined by MTT method to detect the results showed that the GEM processing48h later generations hepatocellular carcinoma HepG2cells significantly inhibited growth, different concentration of GEM average inhibition rate reached45.72%, the cell growth vigor significantly lower than control group (P<0.05). Human hepatocellular carcinoma HepG2cells GEM group of nude mice subcutaneously transplanted tumor tumors had accumulated significantly less than the blank control group (P<0.05).Immunohistochemical detection results showed that the GEM group of nude mouse tumors of the Bel-2gene expression level is lower than the blank control group (P<0.05); GEM group of nude mouse tumors of WWOX gene expression levels higher than the blank control group (P<0.05).Conclusion gemcitabine in human hepatocellular carcinoma HepG2cells can promote apoptosis and inhibit the growth of its, its mechanism may be related to lower the Bel-2and raise the level of gene expression level of WWOX gene expression related.
