Objective: The aim of this study was to observe the expression of MAGE, BAGE and GAGE gene in uterine cervical carcinoma tissue, normal uterine cervical tissue and the cell line of uterine cervical carcinoma,and to evaluate the possibility of using these genes as the targets of specific immunotherapy for uterine cervical carcinoma . The other aim of this study was to investigate the effect of 5-aza-2'-deoxycytidine in proliferation of uterine cervical carcinoma cell line and expression of tumor specific antigen MAGE, BAGE and GAGE .Materials and Methods: The expressions of MAGE-1,MAGE-3,BAGE and GAGE-1/2 in 45 surgically resected cervical squamous cell carcinoma , the Hela cervical cancer derived cell line and 16 of their corresponding normal cervical tissue were studied by a reverse-transcription polymerase chain reaction (RT-PCR), tumor stage by FIGO classification and lymph node involvement. Uterine cervical carcinoma cell(Hela) were treated with 5-aza-2'-deoxycytidine(DAC) in co-culture,the growth curves were observed; MAGE, BAGE and GAGE expression pattern before and after drug treatment was measured by RT-PCR. All data were applied the statisticses software SPSS10.0 to carry on.Results: Expression of MAGE-1, MAGE-3, GAGE-1/2 and BAGE was not observed in normal uterine cervical tissues. MAGE-1 and GAGE-1/2 were expressed of the Hela cell. In contrast, no expression of MAGE-3 and BAGE genes was observed.
