Purposes:Part One: To develop a quick Feulgen stain and try its application in cell nuclei DNA content measurement.Part Two: To evaluate the linearity of photometry for measuring cell image of TIGER cell image analysis system.Part Three: (1)To compare the sensitivity of DNA image cytometry and of conventional cytology in screening of cervical dysplasisa and carcinoma of the uterine cervix.(2)To increase the efficiency and accuracy of cervical cancer screening by using nuclear DNA ploidy analysis.Methods:Part One: Smears of liver cell suspension were obtained from ten healthy mice. The specimens were divided into three groups, stained respectively with quick Feulgen stain, modified one and traditional one. DNA content of intact hepatocyte nuclei was analyzed by TIGER cell image analysis system. The results were compared among different groups.Part Two: Smears of spermatogenic cell suspension from ten healthy mice were stained with quick Feulgen stain. DNA contents of single intact nucleus of the three sorts of testicular spermatogenic cells on the smear were measured with TIGER cell image analysis system. Then the ratio of the nuclear DNA contents among spermatid, spermatogonium and spermiocyte were compared and their extension of AOD was analyzed.Part Three: Each of 100 cervical samples from the women was made into two slides by liquid-based sample preparation.
