The morphology, size, distribution and composition of nanocrystals in urine of lithogenic patients and healthy persons were investigated by means of transmission electron microscopy (TEM), nanoparticle size analyzer, X-ray power diffraction (XRD) and Fourier transform infrared spectroscopy (FT-IR). Most of the nanocrystals in urine of healthy persons are round with a size range from 100 to 300 nm. However, nanocrystals in lithogenic urine are sharp with a size range from several tens to 1000 nm. These results were explained by higher concentration and activity of urinary inhibitors in healthy urine than that in lithogenic urine, the inhibitors in healthy urine can inhibit the growth and aggregation of nanocyrstals. Most of the particles in urine of healthy persons were crystalline material, and the main composition of urinary crystallites in 75% healthy persons was magnesium ammonium phosphate, and that of 12.5% was uric acid, 12.5% was a mixture of calcium oxalate monohydrate (COM) and tricalcium phosphate. However, the main composition of urinary crystallites in 37.5% lithogenic patients were COM, 25% were octacalcium phosphate, 12.5% was a mixture of tricalcium phosphate and COM, 12.5% was tricalcium phosphate, and 12.5% was a mixture of tricalcium phosphate and amorphous materials. The effect of the extract from Tiankui soup on the phase composition and crystal growth of calcium oxalate (CaOxa) were studied by scanning electron microscopy (SEM), XRD and FT-IR. Why the extract from Tiankui soup can inhibit the formation of calcium oxalate stone is that the extract from Tiankui soup can inhibit the nucleation of calcium oxalate, make the COM crystal shape more rounded and inhibit the growth of COM crystals. Moreover, The extract from Tiankui soup can induce the formation of COD crystals.
