ObjectiveTo establish the model of radiation-induced lung injury simulating the radiation methods in clinic. And observe the pathological signs and the dynamic change of cell factor to evaluate the intervention effects of blood-cooling and detoxicating drugs, and do some basic research for mechanism of its preventing and curing function.MethodsAll 160 rats were randomly allocated into control group, irradition-only group,treatment group and drug-only control group . The rats in treatment group and drug-only control group started to be treated with blood-cooling and blood flow-promoting drugs by gastrogavage once daily from 1 week before radiation. The rats in other groups were gastrogavage with 9% NS. The rats in irradition-only group and treatment group were radiated at a dose of 3Gy twice a week in right lung, total 10 times. 8 rats in every group are choosed randomly and sacrificed on the 3rd,5th, 8th,12th and 26th week after first irradiation, We observed the pathological changes and the deposition of collagen by HE staining and Sirius red staining and evaluate the pathology changes with lung injury score .Measuring the different expression of TGF-β ,TNF-α ,IL-6 and bFGF of each group using Image pro plus 5.0.2. after immunohistochemical stain. And statistics the result using SPSS 11.0 software.Results1. T'ypical expression of radiating pneumonia and lung fibrosis are observed in irradition-only group . Through the HE staining and Sirius red staining and evaluate the pathology changes with lung injury score ,we can tell the differentiain the irradition-only group and the control group..2. Slight syndrom of acute inflammation was observed in treatment group. The acute lung injury score is obviously lower than the irradition-only group (P<0.01). In later period of observation, the tiny fibrosis appeared in the irradiated area and the lung fibrosis score is obviously lower than the irradition-only group(P<0.01). The collagen fibers proliferation and the proportionality of type I and III collagen in the treatment group was significantly lower than the irradiation-only group(P<0.01).3. TGF- β , TNF- α, IL-6 and bFGF were expressed higher in the irraditon-only group than the control group (P<0.01). At every observation time point ,the level of these four cell factors in treatment group are obviously lower than the irradiation-only group (P<0.01).Conclusions1. The model of radiation-induced injury of rat lung caused by multiple low dose exposure has lots of merit such as low rate of death,stabilization, good practicability.2. The blood-cooling and blood-flow promoting drugs can lessen the inflammation and delay the progress of pulmonary fibrosis .Through the live animal observation, the lung surface observation and the tissue pathologh observation, the "hot injured lung venation zheng "and "blood retardarce in lung venation zheng" are obviously reduced.3. During the process of generation and progression of radiation-induced lung injury, cell factor interact and change fluctuantly with disease progression .The blood-cooling and blood flow-promoting drugs can reduced the expression of TGF-β , TNF- α, IL-6 and bFGF .This maybe its prevention and treatment mechanism on radiation-induced lung injury.
