Objective: (1) To study the expression of FAK and VEGF in rectal carcinoma, and the relations with biological behavior of rectal carcinoma respectively. (2) To explore the correlation of the expression of FAK and VEGF in human rectal, carcinoma. (3) To investigate the relations between the expression of FAK and VEGF in rectal carcinoma with the prognosis of those patiens with rectal carcinoma. (4) To investigate the relations between the expression of CD31 in the tissue of Rectal carcinoma with the prognosis of patients with Rectal carcinoma.Methods: The expression of FAK and VEGF was examined immunohistochemi cally in normal rectal mucosa, rectal mucosa dysplasia, resected rectal carcinoma, using SABC immunchistochemical method. To investigate the relations between the expression of FAK and VEGF with the prognosis of the patiens. SABC immunchistochemical method was used to detect the expression of CD31 in 86 patients of Rectal carcinoma; To explore the correlation of the expression of FAK and VEGF in human rectal carcinoma.Results:(1) The positive rate of FAK in normal rectal mucosa was 10%, but a relatively high positive rate was found in dysplasia. Of 86 patients tested, 69 (80%) showed positive staining of FAK in the primary lesions. The diference was significant (P< 0.01). The positive rate of those without regional nodal metastasis is 52.9%, significantly lower than those with nodal involvement, which is 98.1 % , The diference was significant (X2=18.97, P<0.01).(2) The positive rate of VEGF in normal rectal mucosa was 13%, but a relatively high positive rate was found in dysplasia. Of 86 patients tested, 48 (56%) showed positive staining of VEGF in the primary lesions. The diference was significant (P< 0.01). The positive rate of those without regional nodal metastasis is 17.6%, significantly lower than those with nodal involvement, which is 80.8%, The diference was significant (X2=33.21, P<0.01).(3) The expression of CD31 in rectal carcinoma has some correlation with the tissue differentiation.CD31 is weakly expressed positive in high-differentiation rectal cancer tissue, is expressed positive in mid-differentiation rectal cancer tissue, is strongly expressed positive in low-differentiation rectal cancer tissue.(4) The expression of FAK has a positive correlation with expression of VEGF and CD31 (P<0.01).(5) Survival rate at 3 and 5 years was 55.1% and 37.7% respectively for positive staining group of FAK, compared to 76.5% and 52.9% respectively for negative staining group(P<0.01).(6) Survival rate at 3 and 5 years was 39.6% and 25.0% respectively for positive staining group of VEGF, compared to 84.2% and 60.5% respectively for negative staining group(P<0.01).(7)Survival rate of the rectal cancer patients for positive staining group of CD31 is significantly lower than those for negative staining group.Conclusions:The expression of FAK in rectal carcinoma has a close correlation with the proliferation, invasion and metastasis of rectal carcinoma. There is a positive correlation between the expression of FAk and VEGF. FAK and VEGF play a key role in the invasion and metastasis of rectal carcinoma and might be a useful tool for prognosis of human rectal carcinoma. The expression of CD31 in rectal carcinoma has some correlation with the prognosis of the patients with rectal carcinoma and may be the index of the prognosis of patients .
