【Objective】To study the expression level and their relationship with clinical pathological parameters of apoptosis-related genes PDCD5 and Survivin in gastric carcinoma,intend to investigate the correlation of the two apoptosis-related genes and their significance in the occurrence and development of gastric cacinoma , so as to provide the valuable evidence for the early clinical diagnosis of gastric cacinoma.【Methods】Expression of PDCD5 and survivin protein in 63 gastric carcinoma tissues,para-carcinoma tissues and 20 normal gastric tissues were detected by Ultrasensitive Streptavidin-Peroxidase immunohistochemistry staining。Expression of PDCD5 and survivin mRNA in 63 gastric carcinoma tissues and 20 normal gastric tissues were detected by Reverse Transcription Polymerase Chain Reaction (RT-PCR).Associate with clinical pathological parameters to analysis the relationship of PDCD5 and Survivin with gastria carcinoma.【Results】1,PDCD5 protein was detected 21 out of 63 cases gastric carcinoma tissue(33.3%),38 out of 63 cases para-carcinoma tissue(60.3%) and 18 out of 20 case normal gastric tissue(90%). PDCD5 mRNA was detected 24 out of 63 cases gastric carcinoma tissue(38.1%),and none of the normal gastric tissue. The expression of PDCD5 mRNA had significant difference between gastric carcinoma tissue and normal gastric tissue(P<0.05).2,The positive rate of Survivin protein in gastric carcinoma tissue was 73.0%(46/63),41.3%(26/63) in para-carcinoma tissue and none Was detected in normal gastric carcinoma. The positive rate of SurvivinmRNA in gastric carcinoma tissue was 79.4%(46/63)and none was detected in normal gastric tissue. The expression of Survivin mRNA had significant difference between gastric carcinoma tissue and normal gastric tissue(P<0.05).3,The expressions of PDCD5 protein and mRNA were correlated with differentiation and lymph node metastasis.There was no relation with age,gender,the depth of invasion and clinical stages.As the decrease of differentiation, the expression rates of PDCD5 protein and mRNA were decreased. The difference between highly and lowly differentiated gastric adenoma had a statistical significance,the neutral differentiated and lowly differentiated gastric adenoma had a statistical significance,but the highly and the neutral differentiated gastric adenoma had no statistical significance(P<0.05). Comparing to without lymph node metastasis,the positive rate of Survivin protein and mRNA in gastric carcinoma tissue with lymph node metastasis was lower.4,The expressions of Survivin protein and mRNA were correlated with differentiation and lymph node metastasis.There was no relation with age,gender,the depth of invasion and clinical stages.As the decrease of differentiation, the expression rates of Survivin protein and mRNA were increased. The difference between highly and lowly differentiated gastric adenoma had a statistical significance,the neutral differentiated and lowly differentiated gastric adenoma had a statistical significance,but the highly and the neutral differentiated gastric adenoma had no statisticalsignificance(P<0.05).Comparing to without lymph node metastasis,the positive rate of Survivin protein and mRNA in gastric carcinoma tissue with lymph node metastasis was higher.5,PDCD5 and Survivin had a negative correlation in gastric carcinoma.【Conclusion】1,Comparing to normal gastric carcinoma tissue, the positive rates of PDCD5 protein and mRNA were dereased.Conclusion can be deduced that promote apoptosis resulted from abnormal expression of PDCD5 had an important significance in the occurance of gastric carcinoma. PDCD5 is a positive regulating factor in apoptosis. Decrease of expression rate has a great prognostication of early diagnosis in gastric carcinoma.2,No Survivin was found in normal gastic tissue,low expression was detected in para-carcinoma tissue,and highly expression was found in gastric carcinoma tissue.Conclusion can be deduced that anti-apoptosis resulted from abnormal expression of Survivin had an important significance in the occurance of gastric carcinoma.3,The expressions of PDCD5 and Survivin were correlated with differentiation and lymph node metastasis.As the dereasing of differentiation and having lymph node metastasis,the expression of PDCD5 was decreased,but the the expression of Survivin was increased. Detecting the expressions of PDCD5 and Survivin may be used to estimate the degree of malignance and prognosis.
