FHIT,Survivin Gene Expression In Human Gastric Carcinoma And Related With Helicobacter Pylori Infection

Objective:To investigate the expression of FHIT,Survivin in human gastric carcinoma and their relationships to clinicopathologic characters and Helicobacter pylori (HP) infection.Methods:Three groups material(gastric carcinoma,corresponding cancer-adjacent tissue and distal tissue) from 62 patients were used.The expression of both protein and mRNA of FHIT,Survivin were detected by immunohistochemistry and reversal transcription polymerase chain reaction(RT-PCR),respectively.The HP infection was detected by boric acid methylene blue staining.Results:(1) Both the protein and mRNA of FHIT expression were significantly different between every two groups(q=2.994～9.176,P＜0.05),and were obviously different in groups of tumor size,invasive depth,TNM stage and lymph node metastasis in gastric carcinoma(Z/t=-3.460～5.725,P＜0.05).(2) Both the protein and mRNA of Survivin expression in carcinomas were high,low in cancer-adjacent tissue,and no expression in distal tissue.There were significant difference between every two groups(q=3.212～16.881,P＜0.05).It was also obviously different in groups of invasive depth,TNM stage and lymph node metastasis in gastric carcinoma(Z/t=-6.457～3.798,P＜0.05).(3) The expression of FHIT,Survivin at both mRNA and protein levels in gastric carcinoma were correlated negatively(r=-0.557,-0.484,P＜0.01).(4) The expression of FHIT,Survivin at both mRNA and protein levels in gastric carcinoma were both correlated to HP infection(r=-0.553～0.393,P＜0.05).The more severe HP infected,the less FHIT expressed.The expression of Survivin was opposed to FHIT.The expression of FHIT mRNA in cancer-adjacent tissue also had a negative correlation with HP infection(r=-0.295,P＜0.05).However,there was no statistically significant difference in the expression of Survivin and HP infection in both cancer-adjacent tissue and distal tissue(r=-0.234～1,P＜0.05).Conclusions:(1) The low expression of FHIT and high expression of Survivin in gastric carcinoma suggest that they may participate in the occurrence of gastric carcinoma.(2) The expression of FHIT,Survivin in gastric carcinoma were both correlated with invasive depth,TNM stage and lymph node metastasis.It indicates that they may play critical roles in the development and progression of gastric carcinoma.(3) The negative correlation between the expression of FHIT and Survivin in gastric carcinoma implies that they may have antagonistic effects in the oncogenesis and progression of gastric carcinoma.(4) HP infection may participate in abnormal expression of FHIT and Survivin in gastric carcinoma.
