Objective:By examining the expression of GRIM-19mRNA in hepatocellular carcinoma tissues and corresponding noncancerous tissues, and GRIM-19protein’s cellular location in liver tissues, as well as the expression level of GRIM-19protein in hepatocellular carcinoma tissues and corresponding noncancerous tissues, in order to investigate the significance and function of GRIM-19in the occurrence and development of hepatocellular carcinoma.Methods:Aseptic technique was adopted to collect40cases specimens of hepatocellular carcinoma. Each group contains a little hepatocellular carcinoma tissues and noncancerous tissues of incisal edge. Expression of GRIM-19mRNA was detected by reverse transcription-polymerase chain reaction in40cases of hepatocellular carcinoma tissues and corresponding noncancerous tissues. The cellular location and the expression of GRIM-19protein was examined by pv two-step immune-histochemical staining technique in40cases of hepatocellular carcinoma and corresponding noncancerous tissues. And western blot was used to detect the expression levels in hepatocellular carcinoma and corresponding noncancerous tissues. The gray values of stripes came from western blot and RT-PCR were analysed by the software of Image J. The chi-square test was used to compare the difference of two rates, while t-test was used to compare the means of two groups. Pearson test was adopted to analyze the relevance. The data were processed by SPSS16.0software. The correlations of GRIM-19to clinicopathologic features were analyzed statistically. Results:1.RT-PCR examination showed that expression of GRIM-19mRNA was lower in hepatocellular carcinoma tissues(0.40±0.31) than in corresponding noncancerous tissues (0.56±0.67), the difference has statistical significance (P<0.05).2.By immunohistochemical detection method,the GRIM-19protein’s positive expression in hepatocellular carcinoma tissues and corresponding noncancerous tissues was presented as varying degree’s tan particles. GRIM-19protein was located in the cytoplasm. The GRIM-19protein’s positive rate was47.5%in hepatocellular carcinoma tissues, while the rate was80%in corresponding noncancerous tissues. The positive rate of GRIM-19protein was obviously lower in hepatocellular carcinoma than in corresponding noncancerous tissues, the difference has statistical significance (χ2=4.57, P<0.05).3.Western blot detection showed that expression of GRIM-19protein was lower in hepatocellular carcinoma tissues (0.31±0.28) than in corresponding noncancerous tissues (0.58±0.21), the difference has statistical significance (P<0.05)4. Statistic analysis revealed as follows:In hepatocellular carcinoma, the expression of GRIM-19mRNA and protein was significantly different between Ⅰ+Ⅱ clinical stages and Ⅲ+Ⅳ clinical stages (0.57±0.38vs0.30±0.20),and so did between groups of portal vein with tumor thrombosis or not(0.04±0.02vs0.44±0.32), Both the differences have statistical significance(P<0.05). The expression of GRIM-19was negatively correlated to AFP value(r=-0.352), the difference has statistical significance (P<0.05). The expression level of GRIM-19was not correlated with age, sex, size of tumor, Liver cirrhosis, metastasis, grading of tumor, the differences have not statistical significance(P<0.05).Conclusions:The hepatocellular carcinoma’s expression of GRIM-19was lower than corresponding noncancerous tissues,which is probably a significant event in the process of hepatocellular carcinoma’s development. GRIM-19may be correlated with the hepatocellular carcinoma’s occurrence, development, invasion and so on.
