Study On Immunity Pharmacodynamics And Anti-tumor Pharmacodynamics Of Thymosin Alpha-1 By Injection

Object:The purpose of this experimentation is to detect the impact of mouse's specificity immune function and non-specificity immune function which are caused by thymosin alpha-1 (Ta1) in injection, and the other purpose is to detect the pharmacodynamics of anti-Lewis lung cancer and S-180 osteosarcoma which are also caused by rh-Ta1.Method:Many methods were used in this experiment, for example macrophage swallowing neutral red, 3-(4,5-dimethylthiazol-2yl)- 2,5-diphenyl tetrazolium bromide (MTT) and ELISA. Using these methods, we can detect the ability of mouse's macrophage phagocytosis, the ability of T,B lympholeukocyte transformation, cytotoxic effector potential caused by natural killer cell(NK) and the level of IL-2. The impact of immune function caused by rh-Ta1 is observed. The examples of Lewis lung cancer and S-180 osteosarcoma are fouded. The probabilities of anti-tumor can be calculated, by injecting different concentration rh-Ta1.Result:1. Rh-Ta1 can improve the ability of mouse's macrophage phagocytosis. The effective is very obviouse in the dose of 0.4 mg/kg and 0.8mg/kg(P<0.05); Rh-Ta1 can improve the ability of T lympholeukocyte transformation. The effective is obviouse in the dose of 0.2 mg/kg and 0.4mg/kg(P<0.001); Rh-Ta1 can improve the abilityof B lympholeukocyte transformation. The effective is obviouse in the dose of 0.4 mg/kg and 0.8mg/kg(P<0.05); It also can improve the level of IL-2. The effective is obviouse in the dose of 0.4mg/kg(P<0.05); NK cell's activity is raised in dose of Rh-Ta1 0.2mg/kg,0.4mg/kg and 0.8mg/kg.2.In the experiment of anti-tumor which includes Lewis lung cancer and S-180 osteosarcoma, the results of anti-tumor are notable.Conclusion:Rh-Ta1 can improve mouse's specificity immune function and non-specificity immune function. It also can restrain the growth of Lewis lung cancer and S-180 osteosarcoma.
