Inhibitive Effect And Mechanism Of Different Mixture Proportions Of Three Kind Of Alkaloid On Tumor Cells

Objective: To select the excellent proportion has the highest inhibitory action on tumor cells in vitro and to study the mechanism of apoptosis.Materials and Methods: MCF-7 cells were treated with matrine, oxmatrine and sophoridine on different concentration. The inhibition ratio of cell reproduction were detected with MTT method. The changes of morphology were observed with microscopy and fluorescence microscopy. Flow cytometer were used to explore the changes of cell cycle.Results:①The inhibition ratio of reduplication of MCF-7 cells was 40%-50% and the concentration of matrine, oxmatrine, sophoridine were during 1.6-3.2mg/ml;②The best excellent proportion is 1:1:1;③The best proportion showed much stronger Inhibitive effect than other proportions.④The best proportion showed much stronger Inhibitive effect than Ma,OM,So;⑤HE staining results showed that microvillus of cells were disappeared, large deciduous apoptosis bodies appeared after being treated with different proportion of Ma,OM,So;⑥Treated 12 lines of tumor cell with 2.4mg/ml mixture alkaloid for 24 hours, the chromatin became red blotches, some nucleus formatted apoptotic body with high coagulation structures and masses of aggregated red apoptosis were observed by fluorescence microscope;⑦the change of the cell cycle was detected with flow cytometry assay and the results showed that the percentage of MCF-7 cells in S stage were decreased and cells in G0/G1 stage were increased after treated with the best proportion mixture alkaloid(1.2mg/ml) for 24hours. The percentage of cells in G0/G1 phase was 69.37%, 65.28%, 64.95% and cells in S phase was 15.00%, 10.63%, 14.94%. But in the group of control, the percentage of cells in G0/G1 phase was 54.06% and cells in S phase was 32.80%Discussion : Apoptosis is a complex process of pathology and physiology, the main way is to start its suicide mechanism by activating endogenic endonuclease. The main mechanism of cure tumor was to evoke tumor cells to apoptosis, such as radiotherapy, chemotherapy, hormone therapy and hot treatments which were used in clinical work. The efficiency of anti-cancer was determined by sensibility of drugs on tumor cells. Matrine is a kind sophora alkaloid which is extracted from a kind of Chinese herbs, a traditional northwest Chinese medicine. All investigations showed that matrine could benefited liver, killed virus, increased leucocyte and improved immunity. The function of matrine was outstanding in many chemotherapy drugs. The inhibition ratio was rise significantly when increased the dose of drug. Furthermore, mixture of alkaloid could induce the twelve cell lines to proceed apoptosis treated with 1.2mg/ml concentration for 24h. Our study also showed that matrine treated tumor cells were induced to apoptosis in vitro, it is maybe the reason of the drug anti-tumor in vitro. Matrine is a promising anti-tumor traditional Chinese drug.
