Objective: Transcatheter hepaticarterial chem oem bolization (TACE) is an important treatment method of HCC, How to better improve the efficacy of TACE and reduce post-treatment recurrence and metastasis has been a hot research field one of HCC. Vascular endothelial growth factor (VEGF) in tumor tissue is the most important angiogenesis factors that not only with tumor angiogenesis, proliferation, but also directly involved in its invasion and metastasis. In addition, Microvessel density (MVD) is the level of influence in patients the important prognostic factor (P = 0.001 2); Many experiments have shown that VEGF and MVD between positive correlation. Now it is known that the tumor proliferation and metastasis depend on angiogenesis. So, VEGF and MVD after TACE changes directly affect the therapeutic effect of TACE. Currently, Anti-angiogenesis research has become a hot spot of Chinese medicine, Particularly through anti-angiogenesis cancer treatment research is particularly prominent.β-elemene is a an anticancer drug that extracted from the rhizome of Chinese temperature Ezhu with a strong effection of killing tumor cells and inhibit tumor growth. Based on the experimental rabbits Vx2 carcinoma model with arterial infusionβ-elemene to further exploreβ-elemene in interventional therapy Intra-arterial administration of the anti-tumor efficacy of angiogenesis.Methods: The rabbits were all transplanted with VX2 carcinoma on liver. Percutaneous transfemoral hepatic arterial catheterization with fixation of the cathether tip inside the feeding vessel was carried out underDSA guidance. 34 rabbits were randomly divided into 5 groups. A group(normal saline 2ml injection ten min), B group(lipiodol 0.5ml via hepatic arterial embolization ten min), C group(β-elemene 50mg one-shot or bolus injection 10min), D group(β-elemene 50mg continuous transarterial infusion 5hour), E group(β-elemene 50mg+lipiodol 0.5ml embolization 10 min). Femoral artery catheterization, The first catheter placed in rabbit liver artery, by different groups were given embolization reperfusion therapy. Anatoming the liver and liver cancer to immunohistochemical detection of the amount of VEGF expression and MVD after the death of rabbits.Streptavidin - biotin - peroxidase (SP) was used to detect the expression of VEGF (vascular endothelial growth factor); Microvessel density (MVD - six) using CD31 monoclonal antibody diluted 1:50 staining was explained, Using SPSS 10.0 software analysis the Group of VEGF and MVD values,P<0.05 was statistically significant。Resuls: Microscopy showed that VEGF-positive cells or plasma membrane staining Thrips uniform. The vascular endothelial cells of tumor are dyeing thrips. Microvessels within the tumor staining were seen mainly in the vicinity of necrotic tissue and tumor on the brink of the brink. MVD within the simply using chemoembolization group compared to other groups has increased significantly, and has significant difference. In theβ-elemene continuous perfusion group and reperfusion group, MVD slightly lower,but no significant statistical difference. MVD slightly higher in lipiodol plusβ-elemene Group, But such trifling differences with the A, C, D groups were not statistically significant. In simply chemoembolization group, VEGF values significantly higher; while VEGF expression scores than saline control group in theβ-elemene infusion, perfusion group and lipiodol plusβ-elemene group VEGF expression scores than saline control group, Upon analysis are all have statistical significance (P <0.05).Conclusion:β-elemene with chemoembolization combining can effectively reduce tumor blood supply , and avoid new tumor angiogenesis. So reduce the postoperative metastasis and tumor recurrence after TACE thereby, and enhance the effectiveness of therapy intervention.
