[Objective]The purpose of this study was to investigate the expression of the mRNA and protein of a new tumor suppressors gene ARHI in the endometriosis, and discuss the invasion mechanism of endometriosis.[Materials and methods]1. Patients were obtained from Dep. Of Gyn. & Obs. in the PLA General Hospital undergoing diagnostic dilatation and curettage, from June to December in 2005. There 23 cases respectively in two groups: 12 of proliferative phase and 11 of secretory phase. Using Northern blot through DNA probe marked by digoxin, detection the expression of ARHI mRNA. Chemiluminescence detection the hybridization. X-ray films exposition in dark room, detected the gray scales.2. Patients were obtained from Dep. Of Gyn. & Obs. in the PLA General Hospital undergoing laparoscopy from August to December in 2005. There 30 cases respectively in 3 groups, including Eu-em and Ec-em groups in EMs and the controls of normal endometrium, derived from the patients undergone surgical operation for the case of infertility or other benign gynecological diseases. Using Ten specimens of ectopic endometrium with endometriosis, ten specimens of eutopic endometrium with endometriosis, and ten control members were investingated by using western blot. Array 3000 scanning apparatus scaned the X-ray films. Using GDS8000 gal-image analysis system detected the gray scales and the ratio.[Results]1. The tumor suppressor gene ARHI mRNA generally express in normal endometrium and endometrosis, none of them express depletion.2. The expression of ARHI mRNA in proliferative phase was higher than thatin secretory phase, but there is not conspicuous difference. And it was not related with age..3. The expression of ARHI protein in eutopic endometrium group was not different with that in eutopic group (P>0.05 );4. The expression of ARHI protein in ectopic endometrium group was extremely significantly higher than that in eutopic group (P<0.001) and that in normal endomerium group (P<0.001).[Conclusion]1. The tumor suppressor gene ARHI mRNA generally express in normal endometrium and endometriosis, and the level of ARHI mRNA was not related with menstrual cycle and age.2. The expression of tumor suppressor gene ARHI was lower in eutopic endometrium with endometriosis than normal endometrium. It maybe related with the implantation in pelvic cavity.3. The expression of tumor suppressor gene ARHI was higher in ectopic endometrium with endometriosis than normal endometrium. It maybe play a role in the pathophysiology and development of endometriosis.4. ARHI always express depletion in ovarian cancer, suggest that it may be related with EMS cancerization, and it is useful to find a new diagnostic and therapeutic target about endometriosis.
