Objective: Compare the biological behavior of epidermal growth factor recepto(rEGFR) andα-catenin (α-cat)both in tissues of colonic cancer and normal colonic tissues.Methods: 20 cases of normal colonic tissues and 68 cases of colonic cancers ,the tissues was from patients who accepted surgical treatments in the Second medical college of Jilin University from January, 2005 to June, 2006 , There are 32 males and 36 females in all patients , the age is between 32-85, the average is 53.2 . No patient accepted Anticancer treatment. The diameters of the tumour is 0.8-12cm, the average is 3.8cm; there are 26 ascending colon cancers , 8 crossing colon cancers; 18 descending colon cancers and 16 sigmoid cancers. there are 48 pipe adenocarcinoma cancers, 10 mammilla adenocarcinoma cancers, 8 mucus adenocarcinoma cancers and 2 non-differentiation adenocarcinoma cancers. 45 cases have lymph nodes metastasis and 23 has no lymph nodes metastasis . 16 cases have distant metastasis (8 liver metastasis , 4 lung metastasis , 3 pelvis metastsis and 1bone metastasis ) and 52 cases have no distant metastasis . According the clinical stage criterion of Dukes: 8 cases in stage A, 15 in stage B, 24 in stage C and 21 in stage D. Tumour cell differentiation: 8 cases are high cell differentiations, 26 cases are medium differentiations and 34 cases are low differentiations. another 20 normal colon tissues which are 5cm away from the edge of tumers . Examine the expressing of EGFR andα-cat in 20 normal tissues and 68 colon cancer tissues by streptavidin /peroxidase(SP) immunohistochmical technique. Based on the pigmentation intensity and the proportion of cells, we classified them into 3 levels by -, +, ++. (1) Grading them by the depth of the color : no staining grade 0 points; staining brown-yellow grade 1 points , staining deep brown grade 2points. (2) Grading them by the proportion of the cell, <30%, 1 points; 30%~71% ,2 points and >71%, 3 points. Pulsing the above two as the final grade of the tissues , and classified by the grade: 0 points is negative (-), 1-3 points is positive (+) and 4-6 points is strong positive (++).Results: There are positive color in the mucous membranes epidermis cell plasm and membranes of the normal colon tissues,especially in the boundary of the cell between cells. Of the 68 colon cancer specimens, there are 17 expressing normal and 51(75%) expressing low.The difference between them is significant (P<0.01). EGFR is hardly expressing in normal colon specimens, but in the cancer specimens, 47(69.1%) appears strong expression, The difference between them is significant (P<0.01). with the gradually increasing of the colon cancer Dukes by stages, the expression ofα-cat is apparently decreased in colon cancer, significant difference appears among the different groups(P<0.05), contrary, the expression of EGFR is increasing apparently, the expressive proportion in stage C and D is higher than in stage A and B apparently (P<0.05). The expressive proportion ofα-cat in colon cancer with the node transferring and beyond transferring is lower than not-transferring specimens(P<0.01). EGFR shows the opposite result (P<0.05). With the descending of the differentiation extent of the colon cancer structure, expression ofα-cat is decreasing gradually and EGFR goes up, the difference is significant(P<0.05).Conclusions: There are positive color in the mucous membranes epidermis cell plasm and membranes of the normal colon,especially in the boundary of the cell between cells. EGFR is hardly expressing in normal colon specimens,with the gradually increasing of the colon cancer by Dukes'stages, the expression ofα-cat is apparently decreased in colon cancer, significant difference appears among different groups, on the contrary, the expression of EGFR is increasing apparently.The expressive proportion ofα-cat in colon cancer with the lymph nodes metastasis and distant metastasis is lower than no metastasis specimens. EGFR shows the opposite result. with the descending of the differentiation extent of the colon cancer structure, expression ofα-cat is decreasing gradually and EGFR goes up, the difference is significant.
